Nathean ATP Torsional Mechanism: Not what we may know about

アブストラクト:
An
unbelievable (almost superhuman?) amount of effort has been
laid into understanding the mechanism of γ-subunit rotation
in F1-ATPase in the hydrolysis mode, especially
by the Tokyo teams, through their ingenious experiments.
Several “protean” hydrolytic schemes have emerged from this
effort: unisite catalysis, bisite catalysis, trisite
catalysis, midway between unisite and bisite catalysis,
midway between bisite and trisite catalysis und so weiter.
Which one do we subscribe to? Be that as it may, ATP
synthesis by ion gradients is experimentally poorly
understood. A primary purpose of my talk is to focus
attention on a parallel (but relatively less known, though
by no means less important!) development of an ingenious
theory of ATP synthesis – the torsional mechanism of energy
transduction and ATP synthesis – which completes ten years
of existence and on which now over 200 journal pages have
been published. This “nathean” view offers a large number of
non-protean mechanistic details of ion gradients-driven ATP
synthesis by F1FO-ATP synthase in the
physiological mode of functioning. The new thinking takes us
beyond Boyer’s and Mitchell’s theories (both of which are
wrong), and what unmitigated faux pas!

There is a remarkable similarity between the three-dimensional covalently bonded networks of boron found in the metal borides and other boron-rich solids and in the boron-hydrogen compounds known as boranes. Surface science studies can reveal some of the properties of the boron clusters found in both types of materials. For example, the icosahedral clusters of boron found in the yttrium boride, YB66, can be observed with scanning tunneling microscopy. Icosahedral clusters of boron and carbon are found in boron carbide and in the carboranes, C2B10H12. Reflection absorption infrared spectroscopy can be used to study the adsorption and dehydrogenation of carborane to form boron carbide thin films.

"Stochastic Gravity: Beyond Semiclassical Gravity"
アブストラクト:
The back-reaction of a classical gravitational field
interacting with quantum matter
fields is described by the semiclassical. Einstein
equations, which have the
expectation value of the quantum fields stress tensor as a
source. The semiclassical theory may be
obtained from the quantum field theory of gravity
interaction with N
matter fields in the large N limit. This theory breaks down
when the fields quantum fluctuations are important.
Stochastic gravity goes beyond the semiclassical limit and
allows for a systematic and selfconsistent description of
the metric fluctuations induced by these quantum
fluctuations. The correlation functions of the metric
fluctuations obtained in stochastic gravity reproduce the
correlation functions in the quantum theory
to leading order in an 1/N expansion. I will describe two
main approaches to stochastic gravity. The first is
axiomatic, the second is based on the open quantum system
paradigm and uses the so called influence functional.
"Structure Formation in the Early Universe"
アブストラクト:
Inflation is known to solve some of the problems of the old
cosmological model, the
so called horizon and flatness problems, which implied some
unnatural initial conditions. However, one of the most
robust and striking features of inflation is that it
provides a natural mechanism to generate the primordial
gravitational fluctuations which are the seeds for cosmic
structure formation. The origin of these fluctuations are
the quantum vacuum fluctuations of the inflation field in the
inflationary period. Inflation predicts a spectrum for the
gravitational fluctuations that is almost scale invariant,
in very good agreement with present observations of large
scale structure. To obtain the spectrum
of
primordial metric perturbation one may use the standard
linear perturbation approach or, alternatively, one may use
the stochastic gravity approach. With the latter it is
possible to fully account for the inflation fluctuations,
beyond the linear approximation.

& kinetics of cooperative cofilin-actin filament
interactions
日程 11月22日 (水) 14:40−16:10
場所 62号館1階 大会議室
紹介教員 石渡信一教授
アブストラクト:
The actin regulatory protein cofilin binds actin filaments
cooperatively and severs them, playing a critical role in
the disassembly and reorganization of the actin
cytoskeleton. Analysis using a nearest neighbor cooperative
lattice binding model indicates that cooperativity is
greater for flexible, non-muscle actin than rigid muscle
actin filaments. The predicted cofilin cluster sizes and
filament binding densities are small at cofilin
concentrations where efficient filament severing is
observed, indicating that a few bound cofilin molecules are
sufficient to destabilize the filament lattice and promote
fragmentation. Ions weaken isolated, noncontiguous cofilin
binding without affecting cooperative filament interactions.
Binding is coupled to the dissociation of ~1.7
thermodynamically bound counterions, which contribute ~40%
of the total cofilin binding free energy (in the presence of
50 mM KCl). The noncontiguous and cooperative binding free
energies are driven entirely by large, positive entropy
changes, consistent with the cofilin-mediated increase in
actin filament microsecond structural dynamics and torsional
flexibility measured using time-resolved phosphorescence
anisotropy. Cooperative binding arises from ~10 fold more
rapid association and ~2-fold slower dissociation. The
results suggest that cofilin-mediated dissociation of actin-associated
ions weakens intersubunit interactions in the actin filament
lattice that enhance cofilin binding site accessibility,
favor cooperative binding and promote filament severing.

Over these last years we have developed theoretical tools which allow us to describe the shape and dynamics of cell compartments controled by actin gels in the presence or not, of molecular motors. We will give example of the two cases. First we will discuss stereocilae and in particular show how one can understand that their length is proportional to their treadmilling velocity and how from the simple shape observation extract useful numbers on the involved forces. Second we will discuss the shape of lamellipodia and the velocity profiles of their actin gel. This will again allow us to extract numbers caracterizing the actin/myosin system in a new and useful way. We will eventully argue that this approach is relevant for the description of cell instabilities including mitosis.

the Higgs Boson
日程 10月16日 (月) 13:30−15:30

紹介教員 山田章一教授
アブストラクト:
The recent measurements of the top quark mass using
data collected by the CDF II detector at the Fermilab
Tevatron proton-antiproton Collider will be presented.
The most accurate measurements employ a novel technique to
constrain the jet energy scale in the lepton+jets
channel, using the observed W boson decay to two jets in the
top quark final states. The implications of this
measurement, the most precise in the world, to
our understanding of the Higgs boson sector will be
summarized.
Research Activities:
My primary interests are the study of pp collisions at the
Collider Detector at Fermilab (CDF), a facility designed,
constructed and operated by a collaboration of physicists
from 40 institutions, and the study of pp collisions using
the ATLAS detector under construction at CERN, Geneva. I am
devoting most of my research effort to the study of the top
quark, the last of the expected fundamental building blocks
predicted by the current “Standard Model” of high energy
physics, and detailed measurements of the properties of the
bottom quark, the partner to the top quark. Having published
conclusive evidence of its existence in 1995, my
collaboration is now involved in a new phase of this
experiment that will increase our top quark candidate sample
by several orders of magnitude. I am also involved in the
development of advanced statistical techniques applied to
the analysis of particle physics data.

I became active in computer graphics 20 years ago, shortly
after
completing my doctoral work in vision/AI. My interest was
kindled by
the surprising realization that graphics modeling had till
then relied
just about exclusively on geometry. It seemed that there was
an
enormous opportunity for physics in graphics, particularly
to produce
animation through simulation. Pioneering work, starting with
the
continuum mechanical modeling of deformable objects, incited
a
revolution of physics-based modeling and animation research,
yielding
innovations such as cloth simulation, which two decades
later are
having a major impact on the motion picture and computer
game
industries. It didn't take me long, however, to realize the
vastly
greater potential of biology-based modeling in graphics. The
confluence of biomechanics, ethology, perceptual and
cognitive
science, this exciting methodology, which appeals to my AI
roots, has
produced virtual animals and even nascently intelligent
virtual
humans. For games, movies, and virtual worlds, this
"artificial life"
paradigm promises autonomous characters that will eventually
be
directable like real actors. This talk will survey the
evolution of my
CG/VR modeling and animation research from physics to
biomechanics and
ultimately to artificial life.
